Dapsone is a sulfone antibiotic. It inhibits dihydropteroate synthase in the bacterial folate pathway, slowing growth and helping treat dermatitis herpetiformis and certain forms of leprosy as part of multidrug regimens.
Dapsone is a sulfone antibiotic used in dermatology and infectious disease. It blocks a key enzyme in folate synthesis, reducing bacterial replication and aiding other therapies in difficult infections.
In practice, dapsone treats dermatitis herpetiformis and is a component of multidrug regimens for leprosy. It may also serve as Pneumocystis jirovecii pneumonia prophylaxis in select patients who cannot take standard sulfa drugs, under specialist supervision.
Do not use if you have a known allergy to dapsone or other sulfone antibiotics. If you have significant anemia or active blood disorders, discuss alternatives with your clinician. G6PD deficiency raises the risk of hemolytic anemia and methemoglobinemia with dapsone and may require testing before starting therapy.
Use with caution in pregnancy or breastfeeding. The drug can cross the placenta and has potential effects on a newborn; benefits and risks should be weighed with a clinician. Impaired liver or kidney function may alter drug levels and necessitate monitoring or dose adjustment.
Common adverse effects include nausea, headaches, and rash. More serious, though less common, events include severe skin reactions, hepatitis, and marked blood dyscrasias. Report unusual fatigue, shortness of breath, chest pain, or yellowing of the skin or eyes promptly.
Tell your clinician about all medications and supplements you take, especially other agents that affect the folate pathway or cause methemoglobinemia. Avoid initiating new therapies without medical guidance, as interactions can raise toxicity or reduce effectiveness.
Common: nausea, loss of appetite, vomiting, headache, fatigue, and mild rash.
Uncommon: dizziness, dark urine, pale skin, abdominal upset, mood changes.
Rare: severe skin reactions, liver enzyme elevations, anemia or other blood cell changes, methemoglobinemia requiring treatment.
Dapsone levels and effects can be altered by rifampin and other liver enzyme inducers. Concomitant use with other folate-pathway inhibitors or agents like trimethoprim-sulfamethoxazole can increase hematologic toxicity and rash risk. Medications or substances that raise methemoglobin, such as certain nitrates, nitrites, or some local anesthetics, may heighten the risk of methemoglobinemia when combined with dapsone. Always inform your healthcare provider about all medicines and supplements you take to manage potential interactions safely.
